How To Create Long-Lasting Botanical Slides?

To prepare permanent slides in botany, three main methods are used: Safranin-Fast Green Combination, Crystal Violet-Erythrosin Combination, and Haematoxylin-Safranin Combination. These methods involve dehydration, clearing, and mounting the samples on the slide for later viewing. Staining and mounting techniques are primarily used for microscopic specimens, such as desmid cell walls, to produce extended depth of focus summary images and three-dimensional (3-D) reconstructions.

To make permanent slides for animals, six main steps are followed: killing, fixing and hardening, staining, dehydration, clearing, and mounting. A thin coat of Canada balsam or DPX is placed on top of the slides, and the solvent evaporates from this. A coverslip is placed on the surface and the slide.

The objective of this research was to evaluate botanical permanent slides using three methods: Tertiary Butyl alcohol (TBA), Xylene-alcohol, and Free hand method. The goal of this process is to maintain the composition of cells, prevent bacteria growth, and prevent decomposition.

A filter paper is placed in a Petri dish, moisten it with water, and replace the lids. The filter paper serves as a mounting medium for making permanent slide preparations of lower cryptogams (epiphytic algae, bryophytes, leaf-associated plants).
